Gimme shelter: three-dimensional architecture of the endoplasmic reticulum, the replication site of grapevine Pinot gris virus.

Una nuova malattia della vite, denominata “Grapevine leaf mottling and deformation” è associata al virus del Pinot gris (GPGV). Utilizzando tecniche molecolari e di immuno-microscopia elettronica abbiamo dimostrato che il virus causa cambiamenti significativi nell’ultrastruttura del reticolo endoplasmico (ER). Sono state eseguite nuove analisi attraverso microscopia elettronica a scansione a fascio ionico focalizzato (FIB-SEM) in tessuti fogliari di vite per dettagliare l’organizzazione tridimensionale dell’ER deformato e il suo rimodellamento dovuto alla replicazione del virus.

Grapevine leaf mottling and deformation is a novel grapevine disease that has been associated with grapevine Pinot gris virus (GPGV). Using molecular and immuno-electron microscopy approaches, we demonstrated that the virus causes significant changes in the endoplasmic reticulum (ER) ultrastructure. Novel analyses using focussed ion-beam scanning electron microscopy (FIB-SEM) were performed in grapevine leaf tissues to detail the three-dimensional organisation of the deformed ER and its remodelling due to virus replication.
